Clinical and biological effects of demethylating agents on solid tumours – A systematic review

•Changes in methylation play a key role in tumour development and therapy resistance.•Demethylating agents are beneficial for a subset of patients with solid tumours.•Decrease in methylation is of limited use to predict clinical response.•Alternative effects e.g. activation of the immune system, are potentially promising.
