•Interstrand crosslink (ICL) repair mechanisms were analysed in Trypanosoma brucei.•Phenotyping T. brucei null lines identified factors that function in ICL repair.•T. brucei ICL repair activities are split across two epistatic groups.•CSB, EXO1 & SNM1 may operate to resolve ICLs encountered throughout the cell cycle.•MRE11, RAD51 & BRCA2 may operate to resolve ICLs encountered during DNA replication.
