•The anatomic changes in primary open-angle glaucoma (POAG) were investigated.•The white mater fibers of POAG were analyzed by a machine learning method.•POAG has anatomic connectivity changes within and beyond of the visual pathway.•FN has potential to be a biomarker for detecting microstructure changes in POAG.
