•A single injection of an adeno-associated viral vector successfully overexpressed neuropeptide S (NPS) in the amygdala.•Prolonged NPS overexpression exerted robust anxiolytic-like effects in rats, but had no effect on depressive-like behavior.•NPS overexpression did not have confounding locomotor or bodyweight effects.
