•Single-molecule magnetic tweezer assay follows eukaryotic transcription initiation.•A 6 bp DNA bubble is generated by the Ssl2 subunit of TFIIH.•Subsequent NTP-dependent transcription expands the bubble to 13 bp.•The kinetics of start-site scanning represents a future mechanistic question.•Correlative fluorescence-tweezers experiments may reveal more moving forward.
