Research on Paclitaxel Microcanti lever Immunosensor

A highly sensitive paclitaxel microcantilever immunosensor was fabricated using sulfhydrated anti- paclitaxel monoclonal antibody immobilized on the gold surface of the microcantilever. An enzyme-linked immunosorbent assay was used to evaluate the activity changes of anti-paclitaxel monoclonal antibody before and after sulfhydrylation, and the bingding activity of sulfhydrated anti-paclitaxel monoclonal antibody immobilized on the gold surface of the microcantilever. The experimental results indicated that although the sulfhydrylation of monoclonal antibody caused loss of 18. 86% bingding activity, sulfhydrated anti-paclitaxel monoclonal antibody immobilized on the gold surface could maintain its paclitaxel binding activity. The paclitaxel microcantilever immunosensor was used to detect different concentrations of paclitaxel solution. The response showed a linear relationaship to the concentration of paclitaxel in the range of 1-100 mu g/L with a detection limit of 0.8 mu g/L (S/N=3). In addition, paclitaxel in human plasma sample was analyzed by microcantilever immunosensor with satisfactory results.
