Automated analysis of rat sperm motility in microchannels

摘要
This work introduces a novel automated analysis of sperm motility in microfluidic channels. The proposed method automates the tracking of sperm as well as estimates its motility parameters. We validate our automated method with manual analysis and ImageJ CASA plugin. Statistical tests show an excellent agreement between sperm trajectories obtained by manual tracking and the proposed automated tracking algorithm. Additionally, three-dimensional (3D) motility parameters such as helical radius and angular frequency of helical motion are evaluated from the two-dimensional (2D) tracked trajectories. We believe that the presented platform has potential to compete with modern CASA method for motility analysis.
