Dendritic Cells Loaded with Heat Shock Protein 70-Peptide to Treat Colon Cancer: An Animal Study

Background: DC-based tumor immunotherapy show great prospects in treating tumors and the treating-effect of therapy mainly depends on the specificity and binding capacity of tumor antigen peptide from tumors. We have modified a feasible way to prepare DC-HSP70/P to provoke immune-reaction. Material and Methods: DCs were cultured from PBMC and HT-29 (the cell line of human rectum and colon gland cancer) was thawed and cultured. HSP70/P had been isolated and purified by DEAE Sepharose (TM) CL-6B Ion Chromatography and ADP-Agarose Affinity Chromatography from HT-29 heated. DC-HSP70/P had been prepared using cultivation method. Then, we observed the anti-tumor effect in vitro and in vivo. Results: DC-HSP70/P had been prepared in success and DC-HSP70/P could suppress tumor proliferation in vitro and inhibit tumor growth, prolong the life-span and enhance the quality life in vivo, which provide a feasible may in further clinical application. Conclusion: We strongly believe that the prospect of DC-HSP70/P will be much more brighter with the study of tumor antigen peptide and the development of Oncomolecular biology.
