ARCUT: Artificial Restriction DNA CUTter: A powerful tool for gene targeting of human genome

We have been developing a chemistry-based Artificial Restriction DNA CUTter (ARCUT) composed of a pair of pseudo-complementary peptide nucleic acids and Ce IV /EDTA complex. This cutter has long (about 16 bp or more) recognition sequence, which enables us to cut huge DNA such as human genome at one target site. Here, we introduce briefly two of our latest studies. The first one demonstrates that ARCUT can cleave human genome at one target site. The second one shows that ARCUT promotes the homologous recombination repair in human cells. These results suggest the possibility of gene targeting of human genome using ARCUT.
