•Dogs with VL show higher expression of PD-1 and its ligands in the spleen cells.•Blocking PD-L1 improves response lymphoprolipherative antigen-dependent in dogs with VL.•IL-10 production decreases in spleen leukocytes from dogs with VL after blocking PD-1.•NO production increases of in dogs with VL after blocking of PD-1 and its ligands.•Splenic parasite load shows positive correlation with PD-1 and PD-L1 on CD21+ cells in dogs with VL.
