•Effectively build adolescent depression model by NMD + CMS, manifested by depression- and anxiety-like behaviors and cognitive dysfunction.•Neuroinflammation reaction mediated by microglial activation was related to adolescent depression induced by NMD + CMS stress.•PI3k/Akt/NF-κB pathway could be involved in the NMD + CMS induced animal model.•Sertraline exerted antidepressive effect in NMD + CMS induced adolescent animal model.
