•Flv1-ΔFlR reveals a monomeric form in contrast with the FDPs “head-to-tail” dimer.•Flv1-ΔFlR pseudo diiron site contains non-canonical basic and neutral residues.•The demetallated Flv1-ΔFlR pseudo diiron site showed an anion moiety.•A functional catalytic site remains to be demonstrated in Synechocystis Flv1.
