Discovery of potent and selective Spleen Tyrosine Kinase inhibitors for the topical treatment of inflammatory skin disease.

•A novel series of SYK inhibitors was identified through fragment and HTS screening.•This was optimised to give selective inhibitors with good cellular potency.•Skin penetration studies were carried out to assess permeability.•A pre-clinical candidate was identified that is currently in Phase I clinical studies.
