•P2Y6 receptor in dorsal spinal cord participate in the development of neuropathic pain after nerve injury.•Suppression of microglia activity will lead to the decreased expression of P2Y6 receptor.•P2Y6 receptor activation is at least in part contributes to nociceptive behavior in CCI- and UDP-treated rats.
