•A novel finding that Exendin-4 promotes VSMC redifferentiation in injured vascular, and thus inhibits neointimal formation.•Exendin-4 promotes VSMC redifferentiation as determined by contractile proteins and cytoskeleton reorganization.•The activation of AMPK/SIRT1/FOXO3a pathways mediate the redifferentiation activity of Exendin-4 in VSMCs.
