•NKCC1 and KCC2 were studied in mice bearing or not β2-V287L, an nAChR subunit linked to sleep-related epilepsy (ADNFLE).•After birth, the KCC2/NKCC1 ratio increased in neocortex, and KCC2 was progressively found in the somatodendritic membranes.•In PFC, KCC2 expression (and the GABAergic switch) was delayed by β2-V287L around P8, but reached higher amounts at P60.•At birth, NKCC1 and KCC2 were abundant in thalamus. By P40, KCC2 declined in RT, and more so in mice bearing β2-V287L.•The regulatory interplay between β2∗ nAChRs and KCC2 in PFC and thalamus may contribute to the pathogenesis of ADNFLE.
