•TAP polymorphisms have no or little effect on peptide transport by TAP using these assays.•Inhibition of TAP by US6, ICP47 and BNLF2a is not affected by polymorphisms of TAP.•Herpesvirus-host coevolution did not result in inhibitor-resistant TAP variants.
