•High leucine concentrations are detrimental to growth upon depletion of 3-MGCoA-H in Trypanosoma brucei..•After depletion of 3-MGCoA-H, mevalonate and FCS restore the growth of PCF and BSF respectively.•Intermediates of leucine catabolism appears to affect the synthesis of mevalonate.
