•Diabetic db/db mice developed a hearing threshold shifts and significant hair cell loss.•MiR-34a plays an important role in diabetes-related hair cell loss by modulating SIRT1 and HIF-1α expression.•MiRNA-34a and HIF-1α are involved in the viability and apoptosis of HEI-OC1 cells induced by high glucose.•MiR-34a contributes to high-glucose-induced hair cell apoptosis via SIRT1/HIF-1α signaling pathway.
