•Neuronal TLR9 antagonism confers protection against excitotoxic death, in vitro.•Neuronal TLR9 antagonism limits excitotoxicity-elicited endoplasmic reticulum stress.•Inhibition of TLR9 in spinal cord astroglia reduces pro-inflammatory cytokine release.•TLR9 antagonism limits astrocyte-elicited neuronal survival in co-cultures.•Thus, a TLR9 antagonist has direct positive and indirect negative effects on neurons.
