•A novel Trichinella cathepsin B-like protease gene, TsCPB2, was characterized.•The recombinant protein TsCPB2 was expressed as an excretory–secretory product.•TsCPB2 antigen induced a mixed Th1/Th2 response and elevated IgE levels in mice.•TsCPB2 antigen induced a protective immunity in Trichinella-infected mice.
