•17β-estradiol (E2) attenuates manganese (Mn)-induced loss of tyrosine hydroxylase (TH) expression in mouse brain.•Tamoxifen (TX) also reverses Mn-induced motor deficits and TH expression.•E2 and TX attenuate Mn-induced reduction of astrocytic glutamate transporters (GLT-1/GLAST).
