•Three serial passages of treated samples improved detection of viable EBOV.•Qiagen viral lysis buffer (AVL) did not fully inactivate EBOV in serum.•Triton X-100 (0.8% Final concentration) did not fully inactivate EBOV in serum.•EBOV in serum was fully inactivated using AVL and Triton X-100 (0.08%) combined.
