One-pot enzymatic glycan remodeling of a therapeutic monoclonal antibody by endoglycosidase S (Endo-S) from Streptococcus pyogenes.

•Endo-S processes various IgG glycoforms and glycan oxazolines with different efficiencies.•Endo-S could transfer hybrid and high-mannose type glycan oxazoline to an intact antibody without product hydrolysis.•Wild-type Endo-S shows complementary activity to Endo-S D233Q mutant in producing different IgG Fc N-glycoforms.•A one-pot IgG Fc glycan remodeling for transforming heterogeneous antibody to homogeneous glycoforms was established.
