•Confirming the over-expression of 17β-HSD7 in primary and progressive Luminal subtype A breast cancer with TCGA cohort.•Suppression of 17β-HSD7 comprehensively modify MCF-7 cell’s proteomes involving in critical biological functions.•GRP78 was found to be a key regulatory protein related to 17β-HSD7 inhibition and effect in MCF-7 cells.
