•We established a hypoxic model of cultured rat neonatal cardiomyocytes.•TRAP1 overexpression improved hypoxia-impaired ATP production.•TRAP1 depletion exacerbated hypoxia-impaired ATP production.•TRAP1 mediated the expression and activity of COXII during hypoxia.•COXII is a downstream effector of TRAP1 in mediating ATP production during hypoxia.
