•PBD molecules bind with the hLig1 enzyme and inhibit DNA joining activity.•They reduce cellular viability and migration of DLD-1 and HepG2 cancer cells.•These hLig1 inhibitors induce apoptosis in DLD-1 and HepG2 cells.
