•The broadly neutralizing mAb 15A7 recognizes the domain 1 of CD4.•The key residues located on the 15A7 binding site overlap with gp120’s.•The mAb 15A7 efficiently blocks the binding of HIV to host cells.•These findings will help in the design of antibody-based drugs for HIV treatment.
