Su1820 Lymphocytic Profile and Cytokines in Patients With Alcoholic Liver Disease

Introduction: The alcohol metabolizing enzymes have several genetic polymorphisms that have been associated with enzyme activity1 and susceptibility to alcoholic liver disease2. Objective: To investigate the frequency of genetic polymorphisms of ADH1B, ALDH2 and CYP2E1 in alcoholics and their relationship with the presence of liver damage. Material and Methods: We evaluated Mexican mestizos subjects: 190 patients (OH) to Liver Clinic of General Hospital of Mexico with alcoholism according to WHO criteria. The control group (CT) consisted of 226 subjects who did consume alcohol of u003c10g/day. Blood samples were taken on a single occasion (10ml) for determination of genetic polymorphisms ADH1B: exon 3, ALDH2: exon 12 and CYP2E1: 5 u0027region (RsaI), intron 6 (DraI) and intron 7 (TaqI). The Hardy-Weinberg equilibrium was achieved. Written informed consent was obtained for each participant. The procedure was approved by the institutional review board. Results: The CT group was AUDIT u003c5, mean age 37±13 years. Patients were classified as alcoholics (47) and cirrhosis (143). Mean age was 46±13 and 49±11 years, respectively. The average grams of alcohol per day for CT were 3±3 while for alcoholics were 307±198 and 315±120 for cirrhotic patients. Average years of consumption in both groups were 28 years. RFLPs: Allele frequencies in significant difference was obtained in the ADH1B * 2 allele (OH 0% vs CT 5% p = 0.001). ALDH2 * 2 allele was a null frequency (0%) in OH and CT groups. For CYP2E1 gene in the promoter region; c1, 16% OH vs 79% CT p = 0.001, for DraI the most frequent allele C with 82% OH and 18% CT with p = 0.001 and for TaqI; A2, with 18% OH vs 13% CT p = 0.51, however these polymorphism showed differences between alcoholics and cirrhotic patients (p = 0.015). Conclusion: Our data reveal genetic patterns in Mexican mestizos that may be in part confer susceptibility to alcohol dependence and risk for alcoholic liver cirrhosis.
