Robust generation and expansion of skeletal muscle progenitors and myocytes from human pluripotent stem cells

•High concentrations of GSK3 inhibitor CHIR99021 induce paraxial mesoderm.•Mixed populations of PAX7-positive cells and skeletal myocytes appear in 6weeks.•Routine passage of differentiating cells allows 1:3 expansion every 2weeks.•Differentiated myogenic populations can be cultured for upwards of 100days.
