Generation and characterization of iPSCs from human embryonic dermal fibroblasts of a healthy donor from Siberian population

Reprogramming of somatic cells into induced pluripotent stem cells (iPSCs) is a powerful method that is necessary for a broad range of studies including substitutive cell therapy and transplantology. Patient specific iPSCs differentiated into relevant cell types represent a valuable model to study pathological mechanisms of inherited diseases at the molecular level. iPSCs from healthy donors are promising for generation of isogenic cell lines harboring mutations in particular genes of interest using genome editing tools. The panel of pluripotent isogenic cell lines and their derivatives represent a good opportunity for pharmacology research such as large-scale drug screening and toxicological studies. In this study, we present iPSCs obtained from human embryonc fibroblasts using episomal vectors. The cell lines express pluripotency markers, demonstrate a stable karyotype 46:XY, and do not contain episome integrations into genome. The cells give rise to derivatives of three germ layers during spontaneous differentiation in vitro and in vivo.
