SDF-1, a CXC-chemokine produced by bone marrow, plays an important role in B-lymphocyte development and trafficking. In this study, we investigated the role of SDF-1 and its receptor, CXCR4, in the chemotactic interaction between CLL cells and bone marrow stromal cells (SC).We observed that CLL cells express surface CXCR4 and migrated in response to recombinant SDF-1. By ELISA, we demonstrated that SC obtained from CLL patients produced significantly more SDF-1 than normal SC leading to increased migration and accumulation of leukemic cells.
