Total Paeony Glycoside Inhibits Tumor Growth and Induces Apoptosis in K562 Cells and in S180 Tumor-Bearing Mice

Total paeony glycoside (TPG) studies on anti-tumor are very little. In this study. It also showed cytotoxic effects against five tumor cell lines and inhibits grow of tumor-bearing mice by establishing a model of mouse sarcoma S180. K562 cells treated with TPG showed typical morphological appearance of apoptosis. The activation of caspase-9 and caspase-3, but not caspase-8, was also demonstrated that the apoptotic signaling triggered by TPG was mediated through the intrinsic mitochondria dependent pathway. The expressions of Bel-2. Bax and Bcl-xL, which were revealed by western blot when K562 cells were treated with TPG. Meanwhile the cell cycle also showed special change with the appearance of GI peak and increase of S cells. TPG could prolong survival time and change the expression of Bcl-2. TPG has anticancer activity and may contribute to the clinical application in cancer prevention and treatment.
