•First GWAS of South Asian population (Punjabi Sikh ancestry) for 25(OH) vitamin D level.•Identification of a novel locus near FOXA2/SSTR4 genes (p=4.47×10−9).•Identification of a suggestive association in IVL gene whose expression in keratinocytes is mediated by calcium and vitamin D response elements (p=1.36×10−6).•Replication of 3 of 5 GWAS loci in Sikhs (GC, CYP2R1 and DAB1) identified previously in Europeans and Hispanics.
