ADVANTAGES OF HIGH CELL DENSITY IN VITRO CULTURES IN DOWN STREAM PROCESSING OF A MONOCLONAL IgG ANTIBODY FOR IN VIVO USE

Abstract This paper analyze the down stream processing parameters of a murine monoclonal IgG antibody for in vivo application. The antibody (ab) was purified from three different starting materials. The down stream procedures were compared in regard to process economy, quality of ab and contamination with LPS. Following starting materials were used: 1. Suspension culture supernatant containing 10% FCS (SC), 2. high cell density bioreactor supernatants containing 10% FCS (TM(FCS)) and 3. protein free high cell density bioreactor supernatants (TM(PF)). We used ammoniumsulphate precipitation (50%). protein A- and G-affinity chromatography (PA, PG) and S-Sepharose Fast Flow ion exchange chromatography (IE) for ab purification. In regard to process economy the advantages of high cell density cultures could be demonstrated. Regarding to LPS contamination IE seems to be the most efficient purification step in case of high cell density bioreactor cultures. In contrast for SC supernatants PA could be shown to be more suitable.
