The Diversification Of Pvyiii. Analysis Of Variability Of P1 Region Of Pvy Using Electrophoresis Of Preformed Dna Heteroduplexes.

The method based on the electrophoretic detection of pre-formed hybrid cDNAs was developed in order to analyze sequence variability of PVY within 394nt fragment of P1 gene. More than 40 PVY isolates from PVYN group were analyzed using this method, In most cases only minor fractions of electrophoretically distinguishable DNA heteroduplexes were found however, in the most isolates of PVYN-Wilga type the mixtures of the major sequence variants were observed, Two of these Wilga clones were sequenced. Both of them differed from the clone PVYNTN (Hungarian isolate) deposited in EMBL database under AC M95491 by nine point mutations, which led to several amino acid changes.
