Polypeptide Synthesis, Solid‐Phase Method

Solid‐phase peptide synthesis (SPPS) has continued to evolve since originally conceived by R.B. Merrifield in 1959. This article is a general introduction into the current state of the art of SPPS. The review covers synthesis strategies, including protecting group combinations, coupling chemistry, monitoring, solid supports, cleavage, and purification. The most troublesome side reactions are also covered. Finally, the prognosis for future developments in SPPS is discussed.
