•p27Kip1 gene expression is regulated positively by MENIN and negatively by MYC and PIM.•Phosphorylation at Thr187 and Ser10 leads of p27Kip1 degradation via SKP2 (nucleus) and KPC (cytoplasm).•PIRH2 degrades p27Kip1 (nucleus and cytoplasm) independent of phosphorylation.•Phosphorylation at Tyr74, Tyr88 or Tyr89 reduces of p27Kip1 inhibitory effect on CDKs.•Phosphorylation at Thr157 or Thr198 increases of p27Kip1 cytoplasmic localization.
