•We validate a specific, precise, and accurate method for indolinone in RHB.•BBB permeation of indolinone was confirmed by in silico and in vitro studies.•Indolinone is not subjected to transporter-meditated efflux.•The calcein uptake assay shows that indolinone does not inhibit P-gp.
