Anticonvulsant activity of pregabalin in the maximal electroshock-induced seizure assay in α2δ1 (R217A) and α2δ2 (R279A) mouse mutants.

•Mutant mice were made expressing α2δ1 R217A and α2δ2 R279A.•Phenytoin activity in maximal electroshock is not affected in these mutants.•Pregabalin reduces seizure severity and latency to seizures in wild type mice.•Activity of pregabalin is not blocked in the α2δ2 mutant, but is in the α2δ1 mutant.•The anticonvulsant action of pregabalin requires binding to α2δ1, not α2δ2.
