•A simple HPLC method is developed, which requires small specimen volume for the measurement of plasma PLP and 4-PA.•The CV% of the method is <7% and the recovery of the added PLP and 4-PA in plasma is about 100%•The concentration of plasma PLP in healthy subjects is associated with plasma 4-PA concentration.
