•Endomorphins presynaptically and postsynaptically produce spinal antinociception.•Both μ1- and μ2-receptors are involved in spinal antinociception of endomorphins.•μ2-Receptors are subdivided into D-Pro2-endomorphin-1-sensitive and -insensitive receptors.•Multiple μ2-receptors differentially contribute to spinal antinociception of endomorphin-1.
