•We studied effects of mPGES-1 deletion on eicosanoids and fatty acids in mice.•mPGES-1 deletion increased PGD2 metabolites and 13-HODE in peritoneal macrophages.•mPGES-1 depletion altered the total lipid fatty acid composition in mouse spleen.
