•Antigen multimerization can markedly enhance antigenicity.•Novel methods for efficient production of multimeric antigens have vaccine relevance.•A multimeric protein vaccine is produced using a peptide spacer and leucine zipper.•Multimeric EBV protein, gp350, is markedly more immunogenic than monomeric gp350.•This method for multimerization of proteins has potential broad applications.
