Isolation and observation of viable sperm cells from the pollens of loquat

In this study, the modified osmotic shock technique and density gradient centrifugation technique were used to isolate and purify the sperm cells of loquat cv. 'Dawuxing'. We observed the vegetative and reproductive cells in pollen tube with the use of DNA specific fluochrome 4',6-diamidino-2- phenylindole(DAPI) and methyl salicylate. The movement and release of sperm cells in pollen tube were observed6 systematically. Living sperm cells were obtained at last, whose density was 4.8 × 106 each milliliter. And the survival rate of them was 54% tested by 0.4% Evans blue.
