Role of Chymase on Generation of AngII and Proliferation of Cultured Human Vascular Smooth Muscle Cells

Purpose To investigate the influence on the cell proliferation and AngⅡ generation by bloc- king chymase path. Methods The human umbilicus artery smooth muscle cells were cultured and divided into five groups:① DMEM group(blank control group); ② AngⅠgroup;③ AngⅠ plus captopril group; ④ AngⅠ plus chymostatin group; ⑤ AngⅠ plus valsatan group.They are designed to block angiotensin-converting enzyme,chymase and AngⅠrespectively.Each group was undergone cell counting, cell proliferation index and AngⅡ concentration in the culture medium tests. Results The cells'numbers were decreased in the drug interfered groups compared with the Ang Ⅰ group (P0.01); the cell proliferation of chymostatin and valsatan group were obviously inhibited(P0.01);the concentration of AngⅡ in chymostatin group was reduced significantly(P0.01), while it was raised in valsatan group(P0.01). Conclusions Chymase plays an important role on generation of AngⅡ and proliferation of cultured human vascular smooth muscle cells.
