Conversion of anti HBs human Fab derived from phage antibody library to whole human IgG molecules

摘要
Objective To convert anti HBs human Fab derived from phage antibody library to eukaryotic expressed whole human IgG molecules. Methods A synthesized human Vκ leader sequence was spliced to anti HBs V H and Vκ by overlap PCR.Eukaryotic expression vectors were constructed and transfected into Sp2/0 or CHO cells. The expression of anti HBs human IgG was tested with ELISA, RT PCR and Western blot. Result Anti HBs human IgG was expressed by transfecting CHO cells with a single vector that contained both light chain and heavy chain genes. Conclusion Fabs obtained from phage antibody library could be expressed in eukaryotic cells using a human Vκ leader sequence.
