Protective effects of polydatin on CCl4-induced injury of primary cultured rat hepatocytec

AIM: To investigate the effects of polydatin (PD) on CCl4-intoxicated primary cultured rat hepatocytes. METHODS: Rat hepatocytes were separated by perfusion with collagenase solution. PD was added into the culture medium after incubating the rat hepatocytes for 24 h, at the same time 10 mol · L- 1 CCl4, was added to induce injury. The level of ALT, malondialdehyde (MDA), reduced glutathione (GSH) and cell viability after cultivation for 6, 12, 24 and 48 h were determined respectively. RESULTS: PD 1 X 10-7 ~ 1 X 10-4 mol · L-1 significantly improved the number of cell viability and prevented the decrease of GSH, ALT and MDA levels. PD 1 X 10-5 mol · L- 1 showed the most effect. CONCLUSION: PD 1 X 10-7 ~ 1 X 10-4 mol · L- 1 could protect primary cultured rat hepatocytes against CCl4-induced injury.
