Influence of t-PA gene transplantation on plasma fibrinolytic activity in mice

We studied the feasibility of enhancing fibrinolytic activity by transplantation of genetic engineering cells. A recombinant expression vector containing human tissue-type plasminogen activator (t-PA) cDNA was constructed and transferred into Chinese hamster lung (CHL) cells. C418-resistant CHL cells were harvested, resuspended in medium containing rat tail collagen solution and incubated again, one ml of the mixture (107/ml) was injected into the subcutaneous tissue and the abdominal cavity of the mice. The t-PA activity in plasma was significantly raised (P < 0.01) and PAI-1 activity was reduced. The results maintained for 3 months. Transplantation of genetic engineering cells might be useful for preventing thrombogenic diseases.
