•EGFRvIII downregulates miR-520-3p which upregulates E2F-1 and ensuing TS in HCC.•5-FU plus mAb CH12 has an additive antitumor effect on EGFRvIII-positive HCC.•CH12-elicited TS suppression contributes to increased sensitivity to 5-FU in HCC.•CH12 is a promising agent to circumvent EGFRvIII-mediated 5-FU-resistance in HCC.
